An amylin drug reached 10.7% weight loss on a different receptor
Not the GLP-1 pathway at all. This one works through leptin, the hormone that tells you that you are full.

Petrelintide, an amylin analog from Zealand Pharma and Roche, produced a 10.7% mean drop in body weight through 42 weeks in the midphase ZUPREME-1 trial, against 1.7% on placebo. Between 88% and 98% of participants reached their target maintenance dose in an escalation of up to 16 weeks, the tolerability case the two companies are leading with.
What makes the number interesting is where it comes from. Petrelintide does not act on the GLP-1 receptor the current obesity injections use; it works on amylin receptors in the brain, which the companies say restores sensitivity to leptin, the satiety hormone. Waist circumference fell 7.9 to 10.8 centimeters against 4.3 on placebo, and a phase 3 program is planned for the second half of 2026.
The trial's primary endpoint was weight loss at 28 weeks, and that data still has not been released; asked when, the companies said only that it is going to another conference. Everything here is a sponsor's presentation rather than a peer-reviewed paper, and analysts who saw the topline in March called the magnitude underwhelming next to the injections already on sale.
Whether a gentler curve is worth a smaller number is the open question, and no chart answers it. That one belongs with a qualified clinician who knows a person's health history.
